Korea Research Institute of Bioscience and Biotechnology has developed a research model of an advanced form of host-symbiotic microorganism by applying in-vitro maturation technology to three-dimensional intestinal organoid differentiated from human pluripotent stem cells.

The research team introduced in-vitro maturation technology to intestinal organoids derived from 3D human pluripotent stem cells to create a research model for intestinal microbial interaction with a microenvironment favorable to engraftment of intestinal microbes.

The mature intestinal organoid contains functional goblet cells that secrete mucin, which is known to be essential for microbes in the gut to attach to the intestinal epithelium and multiply. Through this process, the mucus layer is formed, and the barrier function is enhanced.

As a result, researchers created an environment similar to that of the human intestine, overcoming the limitations of the existing organoid as a research model for intestinal microbes.

The researchers confirmed increased engraftment by injecting lactobacillus, which is a probiotic. They proved the usefulness of organoid matured in vitro to be a research model for studying intestinal microbes.

“The main goal of the organoid research field is to provide high-quality, highly functional human organoid that mimics adults as a testbed for various researches,” said Dr. Son Mi-young of Stem Cell Research Center at KRIBB. “The organoids can be used for researches on treatment and mechanism.”
